Effect of amitriptyline pretreatment on clonidine analgesia was assessed in mouse by acetic acid writhing assay. Amitriptyline potentiated clonidine analgesia and produced a parallel shift of the dose-response curve of the latter, suggesting a common pathway for their analgesic activity. Naloxone failed to reverse their analgesic activity, whereas yohimbine, a selective alpha-2 blocker, completely revesed it. It is suggested that their analgesic activity is not mediated through opioid receptors, but by alpha-2 adrenergic receptors.
