The light absorption in the entire visible range, dissociation constants, and stability of the methemoglobin derivative of mercapoethanol were studied. Hb was isolated from stored human blood using a twofold recrystalization by ammonium sulfate. The color of acid met-Hb changed immediately upon addition of mercaptoethanol. The pK of the mercaptoethanol compound was determined at 406 and 425 nm. The extinction was not constant in any mercaptoethanol-containing solution. It sank slowly in the Soret range. About 15 min after the addition of mercaptoethanol the light absorption increased between 600-690 nm. With its pK value, the mercaptoethanol derivative fits well into the remaining met-Hb complexes.
