To clarify the basic mechanisms of extraocular neuromuscular disorders, experimental formation of neuromuscular unit was performed by co-culture method using the extraocular and brainstem tissues of rat fetuses after individual development in separate tissue culture dish, for which several kinds of technical research was investigated. New neuromuscular junction formation was verified microscopically and demonstrated cinematographically by observing elongation of the nerve fiber, formation of the myotube and finally rhythmic contraction of the muscle fibers innervated by the elongated nerve. Modified injection methods of HRP discharges in human extraocular muscles presenting a case of inferior rectus muscle paralysis by a traffic accident, in which dual innervation by the oculomotor and abducens nerves in the inferior rectus muscle was demonstrated .apprx. 2 wk after the accident. Supranuclear influences on the eye muscle nuclei, especially by the visual stimulation, were demonstrated by the morphological and electrophysiological methods: light deprived effects on cytological changes and their acetylcholine esterase activity changes of the oculomotor nucleus in the mice, influences of electrical stimulation of the optic nerve on the vestibulo-ocular reflex and convergence of neural impulses from the collicular neurons and frontal eye field neurons to the abducens neurons in the cat.
