Changes in fast green-histone amounts with NaCl and HCl treatments in the salivary glands of Bradysia spatitergum

Fast green (FG)-histone values were determined cytophotometrically after NaCl and HCl treatments in nuclei of the salivary glands of the dipteran, B. spatitergum, at the early 4th larval instar. Glandular regions with different levels of secretory activity and DNA content had their FG-histone values compared. A differential decrease in FG-histone values of the various glandular regions with increasing NaCl concentration or acid hydrolysis time is assumed to be related to differential histone removal but also to be affected by histone conformational changes. The most singificant changes in FG-histone values were found in the S2 and S3 regions, known to be involved in intense secretory activity and the largest polyteny levels in these glands.
