Amineptine: clinical considerations on a new antidepressive

Thirty patients (23 outpatients and 7 inpatients, average age 53.13 years) were treated with amineptine and observed for 30 days. Daily dosage was usually 200 mg. Tolerance was optimal in almost all patients. Patients who at the start of treatment were found to have impaired liver or cardiac function were monitored during treatment by clincal and blood chemistry examination and no deterioration of pre-existing damage was observed. Our study confirms the characteristics of amineptine already known from previous trials, i.e. rapidity of action, good tolerability, disinhibiting effect. These pharmacological features would encourage the use of amineptine in depression with inhibition and in patients with heart disease, liver disorders, glaucoma, and prostatic disorders. We paid special attention to the use of this drug in elderly depressed patients with psychomotor inhibition, hypochondriac tendencies (which would be aggravated by a drug with side effects) and in whom other antidepressants would be contraindicated in view of concomitant organic pathologies.
