This review focuses on systemic antifungal agents targeted against opportunistic fungal infections in AIDS patients. Biochemical targets for antifungal drug design, antifungal agents currently on the market (amphotericin B, flucytosine, ketoconazole, itraconazole, fluconazole) or undergoing clinical development (voriconazole, D-0870, SCH-56592, TAK-187, ER-30346, T-8581, genaconazole, saperconazole, SCH-51048, LY-303366, L-743872, aureobasidin A, nikkomycin Z) as well as potential new antifungal agents that have been discovered ( dithiocyclohexadienes, fatty acid derivatives, alkaloids, dihydrobenzonaphthacenequinones, papulacandins, lipopeptides), are discussed.
