Comparison of microscopic & culture findings in the diagnosis of chlamydial urethritis

A comparative evaluation of microscopic examination and culture in the diagnosis of Chlamydia trachomatis urethritis was undertaken. At total of 454 specimens of urethral discharge were stained by I and Giemsa, and cultured on the McCoy cell line. Positive cultures were obtained in 133 patients, whereas cytology was positive only in 14. All the patients positive by cytology were also culture positive. The culture yielded almost 10 times more positive results than cytology. Of the 2 staining methods, i.e., Giemsa and I, Giemsa was superior, as it could detect inclusions in all the 133 positive patients, whereas I staining could detect inclusions only in 22 patients.
