Chronic myelogenous leukemia (CML) may be a conditioned neoplasm because granulopoiesis in CML is, at least in the chronic phase, under regulatory control. Almost all cases of CML revealed leukocytosis and high levels of CFU-C in the peripheral blood as well as the bone marrow. Several abnormalities in the negative feedback control of granulopoiesis in these patients were pointed out, and these defects might cause marked granulocytic hyperplasia. In this study in which colony inhibiting activity (CIA) derived from mature granulocytes was investigated both in patients with CML and in normal subjects, several characteristics (defects) in the negative feedback control of granulopoiesis in CML were seen. CML polymorphonuclear leukocytes (PMN) were defective in the production and/or release of CIA at low concentrations of PMN. CML CFU-C were less sensitive than normal CFU-C to inhibition by low concentrations of CIA. The phenomenon of CIA dissociation was seen at PMN concentrations of over 107 PMN/ml. Although the 1st 2 points had already been elaborated, the 3rd was clarified for the 1st time. A reasonable basis from which the marked granuloid hyperplasia in CML may be explained was provided.
