Immunofluorescence studies showed that the antigen components of calf-pericardium appeared in the ground substance which surrounds the fibers and cells of connective tissue. The soluble proteins in the ground substance can be extracted by Hank's solution or by hyaluronidase. The weight of protein content/g of tissue in each of 2 extracts was almost the same as that being estimated by Peterson's protein assay method. Using isoelectric focusing polyacrylamide gel electrophoresis technique, the soluble proteins were divided into 17 bands. Thymol-sulfuric acid staining and hyaluronic acid digestion showed that the 7th band represents hyaluronic acid. Both the Hank's solution and the hyaluronidase can extract the soluble proteins from the ground substance of calf-pericardium. The extraction seemed more effective when the tissue was treated first by Hank's solution for 12 h and then by hyaluronidase for 5 h, because it was found that the immunofluorescent strength diminished with this treatment. Apparently the tensile strength of the tissue did not decrease after hyaluronidase digestion in comparison with the control.
