A simple method for direct count of blood monocytes

A new, simple method for direct count of blood monocytes, using .alpha.-naphthyl butyrate and hexazotized pararosaniline, was demonstrated. The method is very simple in the staining procedure. The solution can stain blood monocytes only, specifically, clearly and immediately. In comparing this method with the indirect counting method on smear preparations, both results were nearly same (r = 0.99, P < 0.001) when 1500 white blood cells were differentiated on smear preparations. The direct method apparently is reliable.
