Bioavailability of procainamide and disopyramide with and without concomitant administration of an antacid: aluminium phosphate

Ten fasting patients received 750 mg of procainamide orally with water or 11 g of aluminum phosphate mixture in a randomized, single dose, 2-way cross-over study. Plasma concentrations of this antiarrhythmic drug in multiple samples drawn during 12 h after each dose were determined by spectrofluorimetric method. Completeness of procainamide absorption, judged by the area under plasma concentration curve (AUCo .fwdarw. .infin.) decreased with antacid. The peak plasma concentration (Cmax) was reduced but the time to peak plasma concentration was not. In a 2nd study, 10 fasting patients received 200 mg of disopyramide orally with water 11 g of aluminum phosphate mixture with the same methodology. There were no significant differences between treatment means for peak plasma concentration, time to peak plasma concentration and area under plasma concentration curve.
