Isolated working canine heart perfusion apparatus for evaluation of myocardial protection methods

An in vitro model providing with a recirculating circuit using an isolated canine heart and its autogenous blood is described to evaluate the effect of myocardial protection during aortic cross-clamping and to compare merits or demerits of several protective techniques. In this model, a left ventricular function inclusive of left ventricular function curve and myocardial metabolism could be easily measured. This technique may be better in the use of autogenous blood, easy connection of heart with an apparatus and repeated excisions of right ventricular muscles as compared with customary isolated non-working and working heart preparations such as Langendorf apparatus or isolated working rat heart preparation. An isolated canine heart was protected 30-120 min by various myocardial protection methods immediately after heart removal. In the model, an isolated canine heart could work continuously over 3 h without any assistant perfusion and ventricular function and myocardial metabolism could be evaluated.
