Preservation of Toxoplasma gondii by freezing

Suspensions of Toxoplasma gondii were obtained from peritoneal exudate of mice. They were mixed with equal volumes of 10% glycerol solution, 10% dimethyl sulphoxide (DMSO) or 10% DMSO + 10% glycerol (3:1) and then frozen at -70 deg C. They were thawed after 2 to 210 days. The viability of the parasites (as judged by trypanblue exclusion) was 36 to 61% in the different batches, and when the organisms were injected intraperitoneally into mice the mice died in 4.7 to 8 days. After injection of DMSO or DMSO-glycerol mixture the mice died without producing peritoneal exudate, but after the glycerol mixture the mice showed abundant peritoneal exudate with many parasites. It was concluded that glycerol was the best cryo-preservative.
