Serum minocycline kinetics in children

Serum minocycline levels were determined by turbidimetric method using Staphylococcus aureus as the test organism after oral administration (mg/kg) of a loading dose of 2.5 followed by 1.5 in the 12th h in children. The peak serum levels ranged from 0.30-1.15 .mu.g/ml. These were achieved in 41.2% (7 cases) each in the 1st and the 3rd h. In only 17.6% (3 cases), peak values were maintained in the 1st and the 3rd h, indicating peak absorption of the drug from 1-3 h. The mean serum concentration at 6 h was around 50% of the mean peak values. Minocycline level increased to 0.66 .mu.g/ml, 1 h after the second dose. The mean serum level at 24 h was 0.10 .mu.g/ml. The mean and peak serum levels were much lower than in adults. The factors responsible for variability in the serum levels at different time periods with 2 dose regimes are discussed. It is suggested that children would need higher doses of minocycline to achieve levels necessary for higher minimum inhibitory concentration required for its action against resistant organisms.
