The arrangement of immunoglobulin .kappa. genes was studied in hybridomas. Three hybridomas produced by Yeh et al. by fusion between mouse spleen cells and the mouse myeloma NS-1 were compared with NS-1. The DNA of these cell lines were digested with 5 restriction endonucleases (EcoRI, PvuII, BamH-I, HindIII and XbaI), and the sizes of .kappa. gene containing DNA fragments were determined by Southern blot analysis and hybridization with a cloned .kappa. c genes. Subsequently, however, additional rearrangements are possible.
