Maintenance of intact porcine corpora lutea in organ culture

Intact porcine corpora lutea were maintained in organ culture up to 5 days, using a modification of a method previously applied to whole porcine Graafian follicles. The technique employed 37.5% O2 at a pressure of 10 lb/in2. Necrotic areas were rarely seen. The histology of the cultures was well maintained and the cells remained viable; however, they apparently became reduced in size, thereby increasing the extent of the intercellular spaces. Radioimmunoassay of the culture medium for progesterone [P] showed that the glands remained secretively active in the absence of tropic hormones, although the P amount was reduced with increasing time in culture.
