A simple and sensitive procedure was provided for cytochemical staining and assay of .alpha.-glucosidase activity in a single granulocyte cell of chicken bone marrow. Cell suspension spread on a thin 0.05% agar layer over the slide was kept in a half-dried condition and allowed to react with a fluorogenic substrate, 4-methylumbelliferyl-.alpha.-glucoside. The fluorescence formed by cleavage of glucosidic bond was measured by fluorescence microspectrophotometry.
