Pharmacological properties of some androgenic steroid nitrates

Androstanolone nitrate (nitrostanolone), 4-chlorotestosterone nitrate (nichlotest) and testosterone nitrate, were investigated from a pharmacological point of view. All the studied compounds exhibited no significant androgenic-myotrophic, antiandrogenic, estrogenic or progestational activities. They possessed no anti-estrogenic effect. Nitrostanolone had a moderate antigonadotropic activity, whereas other compounds possessed a weak progonadotropic effect. Nitrostanolone failed to affect thyroid function as well as the pituitary-adrenal system. Nichlotest and testosterone nitrate mobilized ACTH. The compounds had no effect upon the uptake of 75L selenium methionine in the proteins of various tissues. Nitrostanolone and nichlotest possessed marked antiinflammatory (antiexsudative) activity. They failed to affect the proliferative processes. Testosterone nitrate had proinflammatory effects. Nichlotest and testosterone nitrate had quite potent coronarodilatatory activity. Nichlotest did not affect the arterial blood pressure, the respiratory movements and the ECG. It lacked any influence upon the cardiovascular changes induced by the i.v. administration of some cholinergic, adrenergic and tryptaminergic agents. It failed to modify some pressor reflexes. All the investigated compounds possessed no visible effects on the CNS, with the exception of nitrostanolone, which exhibited a moderate antipyretic action. Preliminary clinical trials showed that nitrostanolone was a valuable antiinflammatory agent, whereas nichlotest was useful in the treatment of intermediary syndrome.
