Immunological cross-reactivity of anti-sera raised with fluoro dinitro benzene tagged normal human leukocytes against human leukaemic cells

Fluorodinitro benzene (FDNB) tagged normal human leukocytes elicited immune response specific against human leukemic cells without any agglutinating activity against normal leukocytes. Tagged cells with 104 or 105 molecules of FDNB/cell exhibited maximum agglutinating activity against human leukemic cells. With higher or lower proportions of FDNB molecules/cell than this, the anti-leukemic immune response was reduced. Mechanism of action was discussed with special reference to the influence of FDNB-tagging on net cell surface charge, introduction of DNP-hapten moiety on cell surfaces and conformational changes on cell surfaces as a result FDNB induced transformations of hydrophylic groups into hydrophobic groups.
