Subcellular localization of rat testicular hyaluronidase (EC 3.2.1.d) was studied by differential centrifugation. The enzyme was found to be particulate and to sediment with the mitochondrial plus lysosomal fractions. Statistical analysis revealed that Triton X-100 and cetyltrimethylammonium bromide significantly released the bound enzyme. Isatin moderately inhibited the enzyme possibly suggesting that SH group/s are not imperative for catalysis. Oral administration of isatin (200 mg/kg) effected a 153% increase and 59% decrease in rat testicular hyaluronidase activity after 2 and 24 h, respectively.
