A review of current knowledge of the antitumor antibiotic bleomycin, a glycopeptide containing .beta.-lactam rings, produced by II Streptomyces verticillus, is presented. The effect of bleomycin on DNA and RNA, cell sensitivity during specific phases of mitosis and the enzyme (believed to be a hydrolase) inactivating bleomycin are considered. Enzyme activity in cells of mice with planocellular skin cancer is considered. Clinical studies of the effectiveness of bleomycin in humans with various cancers are discussed. During combined use of bleomycin and radiation therapy, an additive effect was observed such that the usual dose of these 2 types of treatment could be decreased and possible toxicity could be avoided. Bleomycin polychemotherapy and future perspectives of bleomycin use are considered.
