Effect of spironolactone on bile formation and liver microsomal enzymic activities in the rat

When given to male rats, spironolactone (15 mg/100 g orally for 4 days) induced a 40% increase in bile flow, which was entirely related to an enhancement of the bile salt-independent fraction. Spironolactone provoked an increase in various microsomal enzymic activities estimated from the 9000 g supernatant fraction. Ethylmorphine demethylase, aminopyrine demethylase and aniline hydroxylase activities were enhanced by 18, 25 and 37%, respectively. Cytochrome P-450 was not modified. UDP-glucuronosyltransferase activity towards 3 substrates, namely phenolphthalein, p-nitrophenol and bilirubin, increased by 38, 45 and 170%, respectively. These effects of spironolactone on bile flow and hepatic microsomal enzymes could account for the attenuation of the pharmacological effect of various drugs given in association with this diuretic.
