Direct spectrophotometric thin-layer chromatography for determining malondialdehyde and beta-formyl pyruvate produced by the periodate oxidation of some deoxy sugars

The periodate-thiobarbituric acid reaction for the quantitative determination of deoxy sugars often gives a mixture of thiobarbituric acid chromogens, namely malondialdehyde and .beta.-formyl pyruvate, which are formed by the periodate oxidation of neutral and acidic deoxy sugars, respectively. The simultaneous determination of these chromogens by spectrophotometric methods is difficult. This paper presents a quantitative TLC method for determining malondialdehyde and .beta.-formyl pyruvate that will be useful as the final stage of analysis of numerous biologically important compounds. The method is applied to Yersinia pseudotuberculosis lipopolysaccharides containing 2-keto-3-deoxyoctonic acid and 3,6-dideoxyhexoses as structural components.
