A simple method of preparing anti-human IgG and its use in quantitation of serum IgG by radial immunodiffusion

A previously described simple method of producing a monospecific anti-human immunoglobin G (IgG) antibody was tested. Human serum has a natural IgG antibody against rabbit red cells. Rabbit red cells were mixed with human .gamma.-globulin, washed and injected i.p. in the same animal on several occasions. A potent anti-IgG resulted. Many attempts were made at standardizing radial immunodiffusion with this antibody and a commercial H chain-specific anti-human IgG. The results with both were comparable. The practical problems encountered, the variables tested and the method which gave reproducible results are described. Sera from 50 normal healthy adults were quantitated for IgG by this method. The mean value obtained was 154 IU/ml and the range was 89-206 IU/ml.
